NXY-059, a neuroprotective agent, reduced disability in acute ischemic stroke patients within 90 days. However, it did not improve neurological function, warranting further research for stroke treatment.
Area of Science:
- Neuroscience
- Clinical Neurology
- Pharmacology
Background:
- NXY-059 is a free-radical-trapping agent demonstrating neuroprotection in animal stroke models.
- Its efficacy in reducing human disability after acute ischemic stroke was investigated.
Purpose of the Study:
- To evaluate the effectiveness of NXY-059 in reducing disability in patients with acute ischemic stroke.
- To assess the impact of NXY-059 on neurological functioning and adverse events.
Main Methods:
- A randomized, double-blind, placebo-controlled trial involving 1722 patients with acute ischemic stroke.
- Patients received a 72-hour infusion of NXY-059 or placebo within 6 hours of stroke onset.
- The primary outcome was disability at 90 days, measured by the modified Rankin scale.
Main Results:
- NXY-059 significantly improved the modified Rankin scale scores compared to placebo (P=0.038).
- No significant improvement was observed in National Institutes of Health Stroke Scale (NIHSS) scores or Barthel index.
- In a subgroup analysis, NXY-059 reduced hemorrhagic transformation and symptomatic intracranial hemorrhage in patients also receiving alteplase.
Conclusions:
- NXY-059 administration within six hours of acute ischemic stroke onset significantly improved disability at 90 days.
- The drug did not significantly improve neurological functioning as measured by NIHSS.
- Further research is required to confirm the benefits of NXY-059 in ischemic stroke treatment.
