    Summary

Guanine dissociation inhibitor (GDI) displacement factor (GDF) is essential for Rab protein membrane delivery. Pure Yip3/PRA1 protein was identified as the key component exhibiting GDF activity, facilitating accurate cellular transport.

    Area of Science:

    • Cell Biology
    • Molecular Biology
    • Protein Trafficking

    Background:

    • Prenylated Rab proteins associate with guanine dissociation inhibitor (GDI) in the cytosol.
    • These Rab-GDI complexes are crucial for the precise delivery of Rab proteins to target membranes.

    Purpose of the Study:

    • To identify the proteinaceous activity on membranes responsible for Rab delivery.
    • To characterize the GDI displacement factor (GDF) and its role in Rab transport.

    Main Methods:

    • Biochemical analysis to determine the properties of GDF activity.
    • Candidate gene approach to identify the protein responsible for GDF activity.
    • Purification of Yip3/PRA1 protein.

    Main Results:

    • GDF activity was found to be membrane-associated with an approximate mass of 25 kDa.
    • Pure Yip3/PRA1 protein demonstrated GDF activity.
    • A method for purifying active Yip3/PRA1 was established.

    Conclusions:

    • Yip3/PRA1 is the protein responsible for GDI displacement factor (GDF) activity.
    • This finding clarifies a key step in the Rab protein membrane delivery pathway.
    • The study provides insights into the purification and analysis of Yip3/PRA1.
