Summary

Lifelong premature ejaculation (PE) is a common male sexual dysfunction. Combining SSRIs with 5-HT(1A) receptor antagonists may offer new on-demand treatments for PE.

Tramadol's Inhibitory Effects on Sexual Behavior: Pharmacological Studies in Serotonin Transporter Knockout Rats.

Frontiers in pharmacology·2018

Area of Science:

  • Neuroscience
  • Urology
  • Pharmacology

Background:

  • Premature ejaculation (PE) is a frequent male sexual dysfunction.
  • Lifelong PE is characterized by rapid ejaculation, often within 2 minutes, starting in adolescence.
  • PE can lead to psychological, sexual, and relationship issues.

Purpose of the Study:

  • Investigate the neurobiological underpinnings of lifelong PE.
  • Evaluate current and potential new pharmacological treatments for PE.
  • Explore novel on-demand therapeutic strategies for PE.

Main Methods:

  • Review of existing literature on PE neurobiology and treatments.
  • Analysis of serotonergic (5-hydroxytryptamine; 5-HT) neurotransmission pathways.
  • Examination of animal studies on SSRI and 5-HT receptor antagonist combinations.

Main Results:

  • Lifelong PE is linked to serotonergic neurotransmission disturbances.
  • Daily SSRIs delay ejaculation but cause side effects.
  • On-demand SSRIs show less efficacy than daily use.
  • Animal studies suggest SSRI and 5-HT(1A) antagonist combinations induce rapid ejaculation delay.

Conclusions:

  • Novel on-demand treatments for PE may involve combining SSRIs with compounds that rapidly enhance 5-HT neurotransmission.
  • Targeting 5-HT(1A) receptors alongside SSRIs shows promise for effective PE management.
  • Further research into combination therapies could lead to improved PE treatments.
