The p53 codon 72 polymorphism is not a risk factor for recurrent pregnancy loss (RPL). This study found no significant difference in p53 gene variants between women with RPL and controls, indicating it does not affect miscarriage risk.
Area of Science:
- Genetics
- Reproductive Medicine
- Molecular Biology
Background:
- Recurrent pregnancy loss (RPL) is linked to altered gene expression.
- The p53 tumor suppressor gene influences apoptosis and angiogenesis.
- A p53 codon 72 polymorphism has been suggested as a potential factor in RPL.
Purpose of the Study:
- To investigate the prevalence of the p53 codon 72 polymorphism in women with RPL.
- To determine if this polymorphism acts as a risk factor for miscarriage.
Main Methods:
- Genotyping analysis of p53 codon 72 variants (Arg72 and Pro72).
- Comparison of genotype and allelic frequencies between 205 women with RPL and 25 controls.
- Polymerase Chain Reaction (PCR) amplification of DNA from buccal swabs.
Main Results:
- No significant differences were found in the frequencies of homozygous (Arg/Arg, Pro/Pro) or heterozygous (Arg/Pro) genotypes between RPL patients and controls.
- Allelic frequencies of the p53 gene showed no significant variation between the study groups.
- Observed allelic frequencies in controls matched previously reported data.
Conclusions:
- The p53 codon 72 polymorphism does not appear to be a susceptibility factor for miscarriage in an unselected population.
- Further research may be needed to explore other genetic or environmental factors in RPL.
