Summary

Mouse zygotic genome activation at the 2-cell stage shows specific transcriptional patterns. These patterns, observed in early embryos and oocytes, suggest a genetic program for multipotent cells.

Area of Science:

  • Developmental Biology
  • Molecular Genetics

Background:

  • The mouse zygotic genome activates at the 2-cell stage.
  • Understanding early embryonic gene expression is crucial for developmental studies.

Purpose of the Study:

  • To characterize the transcriptional activity of microinjected DNA in 2-cell mouse embryos.
  • To investigate the role of specific promoter elements and transactivation in early embryonic gene expression.

Main Methods:

  • Microinjection of DNA constructs into mouse zygotes.
  • Analysis of gene expression using LacZ reporter genes.
  • Qualitative assessment of promoter activity and transactivation.

Main Results:

  • Sp1-sensitive promoters are active in 2-cell embryos.

  • MyoD can activate the acetylcholine receptor alpha-subunit promoter before zygotic genome activation.
  • Conclusions:

    • Early embryonic transcriptional selectivity is present in oocytes and persists post-fertilization.
    • This selectivity shares characteristics with EC cells, suggesting a multipotent cell-specific genetic program.
