Summary

Propolis water solution (PWS) significantly impairs mitochondrial respiration, particularly with succinate substrate. Higher PWS concentrations reduce respiration rates, impacting energy production in heart mitochondria.

Area of Science:

  • Biochemistry
  • Cellular Respiration
  • Mitochondrial Function

Background:

  • Mitochondria are crucial for cellular energy production.
  • Propolis, a natural resin, has various biological activities.
  • Understanding propolis's effects on cellular metabolism is important.

Purpose of the Study:

  • To investigate the impact of propolis water solution (PWS) on rat heart mitochondrial respiration.
  • To determine the dose-dependent effects of PWS on different respiratory substrates.
  • To elucidate potential mechanisms of PWS-induced mitochondrial dysfunction.

Main Methods:

  • Isolated rat heart mitochondria were used.
  • Mitochondrial respiration was measured using NAD-linked (pyruvate + malate), FAD-linked (succinate) substrates, and fatty acids (palmitoyl-L-carnitine).

Related Experiment Videos

  • The effects of varying PWS concentrations (based on phenolic compounds, PC) were assessed.
  • Main Results:

    • PWS at 4 microg mL(-1) PC showed no effect on mitochondrial respiration.
    • PWS at 63 and 125 microg mL(-1) PC significantly decreased basal and maximal respiration rates with succinate (24-70% reduction).
    • Respiration with pyruvate + malate and palmitoyl-L-carnitine was less affected (13-28% reduction).
    • Inhibition of succinate dehydrogenase by esters in PWS may explain succinate oxidation effects.
    • Reduced activity of respiratory chain complexes or ADP/ATP translocator might explain effects with other substrates.

    Conclusions:

    • Propolis water solution exhibits inhibitory effects on mitochondrial respiration in a dose-dependent manner.
    • Succinate oxidation is particularly sensitive to PWS, likely due to succinate dehydrogenase inhibition.
    • PWS may impair cellular energy production by affecting mitochondrial respiratory chain complexes and/or ADP/ATP translocator.
