Summary

Simplified on-farm embryo transfer is now possible for cattle. This study shows that bisected bovine embryos (demi-embryos) can achieve normal pregnancy rates without their zona pellucida, using a new simplified bisection method.

Background:

Purpose of the Study:

  • To describe a simplified method for bovine embryo bisection.
  • To evaluate the viability and pregnancy rates of bisected bovine embryos (demi-embryos) transferred without their zona pellucida.
  • To assess the feasibility of this simplified method for on-farm embryo transfer.

Main Methods:

  • Day 7 bovine embryos were bisected using a microblade and micromanipulator while within the zona pellucida.
  • Bisection was achieved by pinning the embryo between the microblade and the petri dish bottom.
  • Demi-embryos were nonsurgically transferred into synchronized recipients without zonae pellucidae.

Main Results:

  • Pregnancy rates for recipients of demi-embryos were 38% (twin halves) and 45% (single halves).
  • These rates are comparable to the 43% pregnancy rate observed for whole embryo transfers.
  • The study demonstrated that bovine demi-embryos do not require the zona pellucida on Day 7 for successful implantation.

Conclusions:

  • A simplified method for bovine embryo bisection is effective for on-farm embryo transfer.
  • Bovine demi-embryos do not necessitate the zona pellucida for normal pregnancy outcomes when transferred on Day 7.
  • This technique offers a practical approach to enhance embryo utilization in cattle breeding programs.
