We developed Transcript Analysis with Aid of Affinity Capture (TRAC), a novel method for analyzing gene expression. TRAC offers sensitive, PCR-amplified transcript detection without cDNA conversion, making it ideal for routine gene analysis.

Area of Science:

  • Molecular Biology
  • Genomics
  • Biotechnology

Background:

  • Accurate and efficient gene expression analysis is crucial for biological research.
  • Existing methods like microarrays and qPCR have limitations in throughput, sensitivity, or complexity.

Purpose of the Study:

  • To introduce a novel, multiplexed method for focused transcript analysis.
  • To demonstrate the sensitivity, speed, and simplicity of the TRAC method.
  • To compare TRAC with existing quantitative PCR (qPCR) systems.

Main Methods:

  • Transcript Analysis with Aid of Affinity Capture (TRAC) involves hybridizing mRNA targets with detection probes and a biotinylated capture probe.
  • Sandwich hybrids are captured using magnetic streptavidin-coated microparticles, washed, and eluted.

Related Experiment Videos

  • Eluted probes can be PCR amplified and detected via laser-induced fluorescence and capillary electrophoresis.
  • Main Results:

    • TRAC enables simultaneous analysis of tens to hundreds of genes.
    • PCR amplification significantly increases sensitivity, allowing analysis from as little as 2.5 ng of total mRNA.
    • The method demonstrated bias-free amplification for at least 16 PCR cycles.
    • TRAC was successfully adapted to a 96-well format using an automated magnetic particle processor.

    Conclusions:

    • TRAC is a fast, simple, and sensitive method for focused transcript analysis that avoids cDNA conversion.
    • The method is suitable for routine gene expression analysis and complements existing technologies like microarrays and high-density chips.
    • TRAC offers a valuable alternative for researchers needing efficient and sensitive gene expression profiling.
