[Central adrenergic agonist agents as representatives of a new class of nonopiate analgesics]
Abstract:
The problem of nonopiate analgesia is briefly discussed in the historical aspect. The mechanism of the analgesic effect of adrenopositive compounds and its implementation through different populations of alpha-adrenoreceptors at supra- and segmental levels of the brain, the advantages of clopheline over traditional narcotic analgesics are grounded. The data confirming the clinical effectiveness of clopheline in pain syndromes are presented, the perspectives of the practical use of clopheline-like and other adrenopositive agents are discussed.

Adrenergic Agonists: Indirect-Acting Agents
One mechanism involves depleting stored catecholamines by displacing them from synaptic vesicles. These agents, known as "displacers," are transported into vesicles at the expense of noradrenaline. Examples include amphetamine and tyramine, which lack a catechol moiety, resulting in prolonged action, improved oral bioavailability, and...
Adrenergic Agonists: Mixed-Action Agents
Ephedrine and pseudoephedrine lack a catecholamine group, making them less susceptible to degradation by metabolic enzymes. They have increased oral bioavailability and lipophilicity, resulting in a longer duration of action. Their response is reduced by...

Vasopressor or pressor agents: They increase blood pressure and function as cardiac stimulants. Examples include endogenous catecholamines (norepinephrine and dopamine) and synthetic agents (phenylephrine).
