Respiratory syncytial virus (RSV) infection activates Interferon Regulatory Factor (IRF)-3 via reactive oxygen species (ROS) and IKKepsilon kinase. Targeting this pathway may reduce RSV-induced lung inflammation.

Area of Science:

  • Immunology
  • Molecular Biology
  • Virology

Background:

  • Respiratory syncytial virus (RSV) infection triggers chemokine gene expression through transcription factors like Interferon Regulatory Factor (IRF)-3.
  • The precise signaling pathway for RSV-induced IRF-3 activation, particularly the role of reactive oxygen species (ROS), remains incompletely understood.

Purpose of the Study:

  • To investigate the signaling pathway responsible for RSV-induced IRF-3 activation.
  • To determine if intracellular ROS generation mediates this pathway.
  • To identify potential molecular targets for mitigating RSV-induced inflammatory responses.

Main Methods:

  • Utilized alveolar epithelial cells infected with RSV.
  • Assessed the expression and activity of IKKepsilon kinase.

Related Experiment Videos

  • Examined IRF-3 phosphorylation, nuclear translocation, and DNA-binding.
  • Measured RANTES gene transcription, mRNA, and protein synthesis.
  • Employed antioxidants and NAD(P)H oxidase inhibitors to evaluate the role of ROS.
  • Main Results:

    • RSV infection increased IKKepsilon expression and activity.
    • Inhibition of IKKepsilon blocked RSV-induced IRF-3 activation and RANTES gene expression.
    • Antioxidants and NAD(P)H oxidase inhibitors prevented RSV-induced chemokine secretion, IRF-3 phosphorylation, and IKKepsilon induction.
    • ROS generation was confirmed as a critical component in the signaling pathway.

    Conclusions:

    • RSV-induced IRF-3 activation is mediated by ROS generation and IKKepsilon kinase activity.
    • IKKepsilon plays a crucial role in regulating RSV-induced inflammatory gene expression.
    • The ROS-dependent IKKepsilon-IRF-3 pathway represents a novel therapeutic target for managing RSV-associated lung inflammation.
