Background:

  • Rat CD54 is a homologue of human intercellular adhesion molecule-1 (ICAM-1).
  • The rat thyroid cell line FRTL-5 is a well-established model for thyroid research.
  • Lymphocyte adhesion to thyroid cells is implicated in autoimmune thyroiditis pathogenesis.

Purpose of the Study:

  • To investigate the expression and function of rat CD54 in the FRTL-5 cell line.
  • To determine the effect of cytokines and other stimuli on CD54 expression.
  • To assess the role of CD54 in lymphocyte-thyroid cell interactions.

Main Methods:

  • Flow cytometry was used to assess CD54 expression on FRTL-5 cells.
  • FRTL-5 cells were treated with cytokines (interferon-gamma, tumor necrosis factor, interleukin-1) and phorbol 12-myristate 13-acetate.
  • Lymphocyte binding assays were performed using FRTL-5 cells and splenic lymphocytes, with and without blocking antibodies against CD54 and its receptor, lymphocyte function-associated antigen-1 (LFA-1).

Main Results:

  • Basal CD54 expression was observed in approximately 10% of FRTL-5 cells, unaffected by thyrotrophin.
  • Cytokines and phorbol 12-myristate 13-acetate significantly increased CD54 expression.
  • Blocking CD54 reduced lymphocyte binding to FRTL-5 cells, with greater inhibition when combined with anti-LFA-1 antibodies.

Conclusions:

  • Rat thyroid cells (FRTL-5) express CD54, which is upregulated by cytokines.
  • CD54 plays a partial role in mediating lymphocyte adhesion to thyroid cells.
  • These findings suggest CD54-mediated adhesion is important for T cell recognition of thyroid antigens in autoimmune thyroiditis.
