Summary

Lipopolysaccharide (LPS) reduces microglial proliferation by decreasing P2X7 receptor expression. Blocking this purinoreceptor or its signaling pathway inhibits microglial cell cycle progression, highlighting P2X7

Area of Science:

  • Neuroimmunology
  • Cell Biology

Background:

  • Microglia are CNS immune cells that alter their phenotype in response to stimuli.
  • Lipopolysaccharide (LPS) activates microglia, mimicking bacterial infection and causing changes in morphology, proliferation, and purinoreceptor signaling.
  • Activated microglia exhibit decreased P2X7 receptor expression.

Purpose of the Study:

  • To investigate the role of P2X7 receptor down-regulation in LPS-induced microglial antiproliferative effects.
  • To determine if P2X7 receptor signaling is crucial for microglial proliferation.

Main Methods:

  • Utilized P2X7 receptor antagonists (oxidized ATP, KN62, Brilliant Blue G) and ATP-hydrolase apyrase to block P2X7 receptor function.
  • Employed small interfering RNA (siRNA) to down-regulate P2X7 receptor expression in primary microglia and N9 clones.

Related Experiment Videos

  • Performed flow cytometry to analyze cell cycle progression and apoptosis in response to LPS and P2X7 receptor modulation.
  • Main Results:

    • Chronic blockade of the P2X7 receptor significantly reduced microglial proliferation.
    • siRNA-mediated down-regulation of P2X7 receptor expression also decreased microglial proliferation.
    • P2X7 receptor-deficient microglia (N9 clones and primary cells) showed unaffected proliferation with LPS or P2X7 antagonists.
    • LPS and oxidized ATP treatment reversibly decreased cell cycle progression without inducing apoptosis.

    Conclusions:

    • The P2X7 receptor plays a critical role in controlling microglial proliferation.
    • P2X7 receptor signaling supports microglial cell cycle progression.
    • Down-regulation of the P2X7 receptor is a key mechanism underlying the antiproliferative effect of LPS on microglia.
