  • Functional Genomics
  • Enzymology

Background:

  • Identifying novel enzymes is crucial for understanding biological systems but remains a significant challenge in functional genomics.
  • Existing methods often lack the throughput or specificity required for comprehensive enzyme discovery.

Purpose of the Study:

  • To develop and validate a systematic method for discovering novel enzymatic activities.
  • To apply this method for the functional screening of uncharacterized proteins.

Main Methods:

  • Incubation of complex metabolite mixtures with purified candidate proteins.
  • Metabolite profiling using capillary electrophoresis electrospray ionization mass spectrometry (CE-MS) to detect reaction-specific changes.
  • Identification of substrates and products based on altered metabolite levels.

Main Results:

  • The method was successfully validated using known Escherichia coli metabolic enzymes.
  • The approach identified novel phosphotransferase and phosphatase activities for the uncharacterized proteins YbhA and YbiV.
  • These enzymes demonstrated activity towards various sugars and sugar phosphates.

Conclusions:

  • The developed in vitro assay and metabolite profiling approach is an effective strategy for enzyme discovery.
  • This method facilitates the functional characterization of previously unannotated proteins.
  • The approach holds broad applicability for enzyme discovery across diverse biological systems.
