Crotamine, a toxin from rattlesnake venom, distributes primarily to the kidneys and liver after intraperitoneal injection in mice. This study tracked the distribution of both native and detoxified crotamine.
Area of Science:
- Biochemistry
- Toxicology
- Pharmacology
Background:
- Crotamine is a basic polypeptide from Crotalus durissus terrificus venom.
- It induces skeletal muscle spasms and hind limb paralysis in mice.
- Understanding crotamine's distribution is crucial for its potential therapeutic applications and toxicity assessment.
Purpose of the Study:
- To investigate the biodistribution of intraperitoneally injected crotamine in mice.
- To compare the distribution patterns of native and irradiated (detoxified) crotamine.
Main Methods:
- Crotamine purification from Crotalus durissus terrificus venom using FPLC.
- Detoxification of purified crotamine via 2 kGy irradiation.
- Radioiodination of native and irradiated crotamine with Iodine-125.
- Intraperitoneal injection of radiolabeled crotamine into male Swiss mice.
- Quantification of radioactivity in various tissues (blood, spleen, liver, kidneys, brain, lungs, heart, skeletal muscle) at different time points.
Main Results:
- The highest concentrations of radioactivity were detected in the kidneys and liver.
- The lowest radioactivity levels were observed in the brain.
- Distribution patterns were assessed for both native and irradiated crotamine.
Conclusions:
- Crotamine preferentially accumulates in the kidneys and liver following intraperitoneal administration.
- The brain exhibits minimal uptake of crotamine.
- These findings provide insights into crotamine's pharmacokinetic profile and organ-specific distribution.
