Summary

Hereditary copper-associated chronic hepatitis (CACH) in Labrador Retrievers is linked to a genetic defect in copper metabolism. This study found significantly higher liver copper levels in affected dogs, supporting a primary storage disease.

Background:

  • Copper-associated chronic hepatitis (CACH) is a liver disease affecting Labrador Retrievers.
  • The exact cause of CACH in this breed is not fully understood, but a genetic component is suspected.

Purpose of the Study:

  • To investigate the clinical and pathological findings in Labrador Retrievers with CACH.
  • To test the hypothesis that CACH in this breed is caused by a genetic defect in hepatic copper metabolism.

Main Methods:

  • Clinical and pathological data from 15 Labrador Retrievers with CACH were summarized.
  • Liver biopsy specimens were examined histologically, including copper quantification.
  • Eight family members and six unrelated healthy Labrador Retrievers were also examined.

Main Results:

  • Affected dogs showed disproportionate increases in alanine aminotransferase (ALT) relative to alkaline phosphatase (ALP) activity.
  • Histopathology revealed centrolobular copper localization and accumulation associated with liver damage.
  • Mean hepatic copper concentration in affected dogs (1,317 µg/g) was significantly higher than in healthy controls (233 µg/g).

Conclusions:

  • The findings support a hereditary basis for CACH in Labrador Retrievers.
  • A defect in hepatic copper metabolism, likely genetic, is the probable cause of this condition.
