Summary

Interleukin 1 beta and tumor necrosis factor alpha production by monocytes is normal in insulin-dependent diabetes mellitus (IDDM) patients. Differences in monokine responses are linked to TNF-beta gene polymorphism, not HLA class II genes.

Background:

  • Interleukin 1 beta (IL-1 beta) and tumor necrosis factor alpha (TNF-alpha) are implicated in insulin-dependent diabetes mellitus (IDDM) pathogenesis.
  • The human leukocyte antigen (HLA) region genes may regulate monokine production.

Purpose of the Study:

  • To investigate the association between monokine and prostaglandin E2 (PGE2) responses of monocytes (Mo) and HLA-DR types in healthy males.
  • To examine monocyte responses in IDDM patients compared to healthy controls.
  • To explore the influence of TNF-beta gene polymorphism on monokine production.

Main Methods:

  • Monokine assays were performed on Mo cultures stimulated with lipopolysaccharides (LPS).
  • Secretions of IL-1 beta, TNF-alpha, and PGE2 were measured.

    Conclusions:

    • Monocyte IL-1 beta and TNF-alpha production is not significantly different in patients with recent-onset or long-standing IDDM compared to controls.
    • Interindividual variations in monokine responses are likely influenced by TNF-beta gene polymorphism rather than HLA class II genes.
    • These findings may contribute to understanding the association between HLA haplotypes and autoimmune diseases.
