Interaction between GNB3 C825T and ACE I/D polymorphisms in essential hypertension in Koreans

Essential hypertension (EH) risk is linked to gene interactions. The G-protein beta3-subunit (GNB3) 825T allele and angiotensin I-converting enzyme (ACE) D allele interact, increasing EH predisposition in Koreans.

Background:
- Essential hypertension (EH) is a polygenic disease where gene-gene interactions are critical for understanding genetic influences.
- The G-protein beta3-subunit (GNB3) C825T polymorphism and the angiotensin I-converting enzyme (ACE) insertion/deletion (I/D) polymorphism are candidates for EH development.
Purpose of the Study:
- To investigate the association between GNB3 C825T and ACE I/D polymorphisms and their interaction in the development of EH in the Korean population.
Main Methods:
- A case-control study involving 688 hypertensive and 924 normotensive Korean subjects.
- Genotyping of GNB3 C825T and ACE I/D polymorphisms using polymerase chain reaction (PCR) and PCR-restriction fragment length polymorphism.
- Logistic regression analysis to assess associations and gene-gene interactions.
Main Results:
- Neither GNB3 C825T nor ACE I/D polymorphisms alone were significantly associated with EH status.
- The GNB3 825T allele showed a positive association with EH in males (OR, 1.459).
- A significant gene-gene interaction was found between the GNB3 825T allele and the ACE D allele (P<0.05), with a higher EH risk in carriers (OR, 1.490).
Conclusions:
- The GNB3 C825T and ACE I/D polymorphisms individually do not strongly predict EH in Koreans.
- A significant interaction between GNB3 825T and ACE D alleles contributes to EH predisposition in the Korean population.
