    Background:

    • Japanese encephalitis is a severe neurological disease caused by a virus.
    • The cellular response within the brain during viral encephalitis is not fully understood.

    Purpose of the Study:

    • To identify the origin of inflammatory cells in the brain during experimental Japanese encephalitis.
    • To characterize the morphology of these inflammatory cells.

    Main Methods:

    • Adult mice were injected with 3H-thymidine to label circulating blood cells.
    • Mice received an intracerebral injection of Japanese encephalitis virus.
    • Brains were analyzed using light and electron microscopic autoradiography on days 3, 4, and 5 post-inoculation.

    Main Results:

    • Inflammatory cells in the brain parenchyma and perivascular spaces were derived from circulating mononuclear leukocytes.
    • These cells adopted a "rod cell" morphology.
    • These "rod cells" were the primary constituents of brain "glial nodules".

    Conclusions:

    • Circulating mononuclear leukocytes are the source of inflammatory cells in the acute phase of experimental Japanese encephalitis.
    • "Rod cells" observed in the brain are derived from these leukocytes and form glial nodules.
