Area of Science:

  • Biotechnology
  • Materials Science
  • Immunology

Background:

  • Antibody microarrays are crucial for high-throughput diagnostics.
  • Antibody stability during printing and storage is a key challenge.
  • Optimizing print buffers can enhance microarray performance.

Purpose of the Study:

  • To evaluate hydroxylated additives for stabilizing antibodies on microarray slides.
  • To determine the optimal additive for antibody print buffers.
  • To assess the long-term stability and performance of stabilized antibodies.

Main Methods:

  • Various hydroxylated additives were tested in antibody print buffers.
  • Polymer-coated microarray slides were used for antibody printing.
  • Spot morphology, antibody distribution, fluorescence intensity, and analyte capture activity were analyzed.

Main Results:

  • Polyvinyl alcohol (PVA) addition resulted in regular spot morphologies and homogeneous antibody distribution.
  • PVA-treated microarrays showed uniform fluorescence intensity.
  • Analyte capture activity for anti-human IL-1beta, IL-4, and TNFalpha was improved and maintained for 1 month at 4°C.

Conclusions:

  • Polyvinyl alcohol is an effective additive for stabilizing antibodies in print buffers.
  • PVA enhances the quality and performance of antibody microarrays.
  • Stabilized antibody microarrays demonstrate robust performance for detecting biomarkers.
