Summary

Microwave assistance significantly speeds up the preparation of polyethylene glycol (PEG)-Cibacron blue and Sepharose-Cibacron blue affinity materials. These microwave-assisted methods offer comparable enzyme purification performance to conventional techniques.

  • Purification of alcohol dehydrogenase and EcoRI was performed using both conventionally and microwave-assisted prepared materials.

    • Sepharose CL-4B-Cibacron blue 3GA preparation time was shortened to 21 minutes (from 3.5 hours) using microwave assistance.
    • Enzyme purification performance for alcohol dehydrogenase and EcoRI was comparable between materials prepared by conventional and microwave-assisted methods.

    Conclusions:

    • Microwave assistance offers a significantly faster and efficient alternative for synthesizing key affinity materials.
    • The rapid preparation methods do not compromise the effectiveness of affinity macroligands and media in enzyme purification.
    • This approach has the potential to streamline biochemical purification processes.
