Structure and Function of Platelets

The cell fragments known as platelets are disc-shaped, with an average diameter of about 3 μm and a thickness of roughly 1 μm. They play a crucial role in the body's vascular clotting system, which also involves plasma proteins, blood cells, and blood vessel tissues.

Background:

Purpose of the Study:

  • Evaluate ADVIA 120 and Thromboelastogram (TEG) for assessing stored platelet changes.
  • Determine correlation between TEG results and other common platelet measures.

Main Methods:

  • Platelet concentrates tested at 1 and 5 days of storage.
  • Assessed platelet count, aggregation, hypotonic shock response (HSR), Kunicki morphology score (KMS), CD62 expression, platelet density, granule content (ADVIA 120), and clot formation kinetics (TEG).

Main Results:

  • Platelet count and HSR remained stable; aggregation decreased by 50% by day 5.
  • Kunicki morphology score (KMS) dropped significantly; mean platelet component (MPC) fell below 200 g/L.
  • Slight platelet activation observed; clot formation rate decreased, with TEG results correlating well with KMS.

Conclusions:

  • Standard in vitro platelet function tests show alterations during storage.
  • A MPC < 200 g/L correlated with KMS < 200.
  • TEG and KMS measurements may predict platelet survival and recovery.
