Summary

Organic phosphates like 2,3-DPG and inositol hexaphosphate (IP6) shift the oxygen-binding curve of sheep hemoglobin. Both compounds increase P50, indicating reduced oxygen affinity, with IP6 having a more pronounced effect.

Area of Science:

  • Biochemistry
  • Physiology

Background:

  • Hemoglobin's oxygen affinity is modulated by organic phosphates.
  • 2,3-diphosphoglycerate (2,3-DPG) and inositol hexaphosphate (IP6) are key regulators.

Purpose of the Study:

  • To investigate the impact of 2,3-DPG and IP6 on the oxygen-equilibrium curve (OEC) of different sheep hemoglobin types (A, B, and F).

Main Methods:

  • Thin film method (HEM-O-SCAN) was employed.
  • Experiments were conducted in bis-tris-saline buffer, excluding CO2.
  • Hemoglobin concentration was reduced to mimic in vivo conditions.

Main Results:

  • Both IP6 and 2,3-DPG caused a rightward shift in the OEC, increasing P50.
  • IP6 increased P50 by a factor of 1.58-1.84, while 2,3-DPG increased it by 1.41-1.59.

Related Experiment Videos

  • 2,3-DPG did not significantly alter the Hill n (cooperativity index), whereas IP6 decreased it.
  • Conclusions:

    • Organic phosphates 2,3-DPG and IP6 reduce hemoglobin-oxygen affinity in sheep.
    • IP6 demonstrates a stronger effect on P50 and cooperativity compared to 2,3-DPG.
