Science advances·2025

Automated optical coherence tomography (OCT) analysis may underestimate retinal thickness and height in patients with central serous chorioretinopathy (CSC) or neovascular age-related macular degeneration (AMD). Always use original OCT images for accurate measurements in these conditions.

Area of Science:

  • Ophthalmology
  • Medical Imaging
  • Retinal Diseases

Background:

  • Optical coherence tomography (OCT) is crucial for assessing retinal structure.
  • Automated analysis of OCT images aims to standardize measurements like retinal thickness and height.
  • Accurate measurements are vital for diagnosing and monitoring retinal conditions.

Observation:

  • This study compared original OCT scans with images after automated retinal thickness analysis.
  • Retinal thickness and height were measured in normal eyes and eyes with various macular conditions, including neovascular age-related macular degeneration (AMD) and central serous chorioretinopathy (CSC).

Findings:

  • Automated analysis showed no significant difference in retinal thickness for normal eyes, macular holes, or non-AMD related macular edema.

Related Experiment Videos

  • However, automated analysis significantly underestimated retinal thickness and height in eyes with CSC and neovascular AMD compared to original scans.
  • Bland-Altman plots indicated substantial mean differences in measurements for CSC and neovascular AMD groups.
  • Implications:

    • Retinal thickness and height measurements may be underestimated in CSC and neovascular AMD when using analyzed Stratus OCT images.
    • It is recommended to use original scanned OCT images for accurate retinal thickness and height measurements in patients with CSC or neovascular AMD.
    • This finding impacts the clinical interpretation of OCT data and patient management for these specific conditions.
