We developed dual-pump coherent anti-Stokes-Raman scattering (dual-CARS) microscopy for simultaneous imaging of two species. This advanced technique effectively removes background noise, enhancing image contrast and enabling clear visualization of molecular vibrations.
Area of Science:
- Biomedical optics
- Molecular imaging
- Microscopy techniques
Background:
- Coherent anti-Stokes-Raman scattering (CARS) microscopy enables label-free vibrational imaging.
- Simultaneous imaging of multiple species and nonresonant background suppression remain challenges in CARS microscopy.
Purpose of the Study:
- To introduce a novel dual-pump coherent anti-Stokes-Raman scattering (dual-CARS) microscopy technique.
- To enable simultaneous imaging of two distinct molecular vibrations and suppress nonresonant background signals.
Main Methods:
- Utilizing three synchronized laser pulses to probe two different molecular vibrations.
- Implementing a dual-pump excitation scheme within the CARS framework.
- Applying the technique to image lipid mixtures and biological samples.
Main Results:
- Successfully imaged a mixture of nondeuterated and deuterated lipids, distinguishing individual components and their organization.
- Demonstrated simultaneous imaging of two species with different vibrational frequencies.
- Achieved significant enhancement in image contrast by suppressing nonresonant background.
- Visualized lipid stores in living Caenorhabditis elegans nematodes with improved clarity.
Conclusions:
- Dual-CARS microscopy is a powerful tool for simultaneous, label-free imaging of multiple molecular species.
- The technique offers superior image contrast through effective nonresonant background suppression.
- Dual-CARS microscopy has significant potential for biological and chemical imaging applications, particularly in complex systems like lipid organization.

Raman Spectroscopy Instrumentation: Overview
The monochromatic laser source, typically using visible or near-infrared radiation, generates a highly focused beam of light. This light interacts with the molecules of the sample, scattering some of the light. Liquid and gaseous samples are usually tested in ordinary glass capillaries, while solids can be analyzed as powders packed in capillaries or as potassium...
Phase Contrast and Differential Interference Contrast Microscopy
In-phase-contrast microscopes, interference between light directly passing through a cell and light refracted by cellular components is used to create high-contrast, high-resolution images without staining. It is the oldest and simplest type of microscope that creates an image by altering the wavelengths of light rays passing through the specimen. Altered wavelength paths are created using an annular stop in the condenser. The annular stop produces a hollow cone of...
