Cyclic GMP and lectin-induced lymphocyte activation.

Summary

This study found no consistent increase in cyclic guanosine monophosphate (cGMP) in human lymphocytes stimulated by plant lectins. Cyclic adenosine monophosphate (cAMP) levels did increase, questioning cGMP's role as a second messenger.

Background:

Mitogenic plant lectins like phytohemagglutinin and concanavalin A are used to stimulate human peripheral lymphocytes.
Cyclic guanosine monophosphate (cGMP) has been proposed as a second messenger in lymphocyte activation.

Purpose of the Study:

To investigate the role of intracellular cyclic guanosine monophosphate (cGMP) and cyclic adenosine monophosphate (cAMP) in human peripheral lymphocytes stimulated by mitogenic plant lectins.
To determine if cGMP acts as a second messenger in this cellular response.

Main Methods:

Purified human peripheral lymphocytes were incubated with phytohemagglutinin and concanavalin A.
Intracellular cGMP and cAMP levels were measured using radioimmunoassay after various isolation and purification techniques.
Lymphocyte activation was assessed by 45Ca uptake and 3H thymidine incorporation.

Main Results:

No consistent increases in intracellular cGMP were detected under any experimental conditions.
Exogenous cGMP was measurable, confirming assay validity.
Small, statistically significant increases in intracellular cAMP were observed.
Other lymphocyte activation parameters, including 45Ca uptake and 3H thymidine incorporation, remained unimpaired.

Conclusions:

The data challenge the established concept of cGMP as the primary second messenger in lectin-stimulated human peripheral lymphocytes.
Cyclic adenosine monophosphate (cAMP) shows a consistent increase, suggesting a potentially different or additional role in lymphocyte activation.
Further research is needed to elucidate the precise signaling pathways involved in lymphocyte activation by mitogens.
