Disabled-2 (Dab2) protein is crucial for embryonic blood vessel formation. It regulates vascular endothelial growth factor (VEGF) induction by activin-like signaling during early development in Xenopus.
Area of Science:
- Developmental Biology
- Molecular Biology
- Cell Biology
Background:
- Embryonic vascular system formation mechanisms are not fully understood.
- Disabled-2 (Dab2), a cytosolic adaptor protein, is investigated for its role in blood vessel development.
Purpose of the Study:
- To elucidate the role of Disabled-2 (Dab2) in embryonic vascular development.
- To investigate the molecular mechanisms by which Dab2 influences angiogenesis in Xenopus.
Main Methods:
- Utilized antisense morpholino oligonucleotide (MO)-mediated knockdown and overexpression of Xenopus Disabled-2 (XDab2).
- Performed functional assays in Xenopus animal caps to assess gene induction.
- Analyzed intersomitic vein (ISV) formation and vascular endothelial growth factor (VEGF) expression.
Main Results:
- XDab2 localizes to blood vessels, including intersomitic veins (ISV), in early Xenopus embryos.
- Both XDab2 knockdown and overexpression inhibited ISV formation.
- XDab2 depletion blocked activin-like signal-induced VEGF expression but not other target genes.
- VEGF coexpression rescued ISV formation in XDab2-depleted embryos.
Conclusions:
- XDab2 plays a critical role in embryonic angiogenesis in Xenopus.
- XDab2 mediates VEGF induction by activin-like signaling during early development.
- This study highlights a novel regulatory pathway in vascular development.
