Leukaemic stem cells in acute myeloid leukaemia (AML) patients with CD33-positive AML express CD33. This finding supports using CD33-targeting antibodies for novel AML therapies.
Area of Science:
- Hematology
- Immunology
- Cancer Biology
Background:
- CD33 (Siglec-3) is a key target for antibody therapies in acute myeloid leukaemia (AML).
- In normal blood development, CD33 is expressed late, not on primitive stem cells.
- This study investigated CD33 expression on leukaemic stem cells in AML.
Purpose of the Study:
- To determine if leukaemic stem cells in AML patients express CD33.
- To evaluate the potential of CD33 as a target for AML treatment.
Main Methods:
- Utilized multicolour staining and cell sorting to isolate leukaemic progenitors (CD34+/CD38-/CD123+).
- Analyzed CD33 mRNA expression in purified leukaemic stem cells using RT-PCR.
- Confirmed AML stem cell populations via NOD/SCID mouse repopulation experiments.
Main Results:
- Leukaemic progenitors from CD33-positive AML patients expressed CD33.
- AML stem cells in CD33-positive AML reside within the CD33+ subpopulation.
- CD33 mRNA was detected in highly purified leukaemic stem cells from CD33+ AML patients.
- CD33 was absent on normal hematopoietic stem cells and leukaemic stem cells in CD33-negative AML.
Conclusions:
- Leukaemic stem cells in CD33-positive AML patients demonstrably express CD33.
- This supports the development of novel therapeutic strategies targeting CD33 in AML.
