Summary

Functional suppressor cells are not responsible for the inability to transfer experimental hypersensitivity pneumonitis (EHP). Cell populations capable and incapable of transferring EHP did not differ in T and B cell composition or size.

Background:

  • Experimental hypersensitivity pneumonitis (EHP) can be transferred via lymph node cells (LNC) cultured with antigen.
  • The presence of functional suppressor cells in EHP transfer systems has not been fully elucidated.

Purpose of the Study:

  • To investigate the role of functional suppressor cells in the transfer of EHP.
  • To characterize the cellular composition of EHP-transfer competent and incompetent cell populations.

Main Methods:

  • Guinea pigs were sensitized and challenged with Micropolyspora faeni (M. faeni).
  • Lymph node cells (LNC) were cultured with M. faeni antigen and transferred to recipients.
  • Flow cytometry and monoclonal antibodies were used to analyze cell populations (T cells, surface Ig+ cells, large cells).

Main Results:

  • LNC cultured with M. faeni antigen from 2-week donors transferred EHP, indicated by increased pulmonary abnormalities.
  • Cell populations from 0-micrograms/mL M. faeni 2-week donors or 10-micrograms/mL M. faeni 8-week donors were incapable of transferring EHP.
  • The addition of incompetent cell populations did not inhibit EHP transfer by competent populations.

Conclusions:

  • The inability to transfer EHP is not due to the presence of functional suppressor cells.
  • Differences in EHP transfer capability do not correlate with variations in cell size distribution or T and B cell composition.
