Summary

Arsenic exposure damages peripheral nerves by altering key proteins, particularly neurofilaments, in rat sciatic nerves. This study reveals the underlying molecular mechanism of arsenic-induced neurotoxicity.

Background:

Arsenic exposure is linked to cancers and neurological disorders like peripheral neuropathy.
Clinical and electrophysiological studies confirm arsenic's peripheral neurotoxicity, showing reduced nerve conduction velocities.
The precise mechanism of arsenic's impact on the peripheral nervous system (PNS) remains largely undescribed.

Purpose of the Study:

To investigate the molecular mechanisms of arsenic toxicity on the peripheral nervous system.
To analyze the protein composition changes in the sciatic nerve following arsenite exposure in rats.

Main Methods:

Male Wistar rats were administered a single intravenous dose of arsenite.
Sciatic nerves were excised post-sacrifice for protein composition analysis.
Western blot analysis was used to quantify neurofilament and fibroblast proteins.

Main Results:

Arsenite exposure led to the disappearance of neurofilament and fibroblast proteins in rat sciatic nerves.
A dose-dependent decrease in neurofilament-M and -L proteins was observed with increasing arsenite dosage.
Changes in cytoskeletal protein composition, indicated by reduced Western blot signals, were evident.

Conclusions:

Arsenic exposure significantly alters protein composition in the rat sciatic nerve, with a notable impact on neurofilaments.
These protein alterations, particularly in the cytoskeleton, are likely responsible for the observed in vivo neurotoxic effects of arsenic.
