Summary

Endothelin-1 (ET-1) and big endothelin (big ET) are found in amniotic fluid (AF). Concentrations of both ET-1 and big ET significantly increase in AF from mid-trimester to full-term pregnancy.

Background:

  • Endothelin-1 (ET-1) is present in both circulation and amniotic fluid (AF).
  • Understanding ET-1 and its precursor, big endothelin (big ET), in AF is crucial for pregnancy research.

Purpose of the Study:

  • To quantify ET-1 and big ET concentrations in plasma and AF.
  • To identify the molecular forms of endothelin in human AF using advanced techniques.

Main Methods:

  • Radioimmunoassay (RIA) was used to measure ET-1 and big ET levels.
  • High-performance liquid chromatography (HPLC) coupled with RIA characterized endothelin molecular forms in AF.
  • C18 cartridges were used for sample extraction from plasma and AF.

Main Results:

  • Both ET-1 and big ET were detected in plasma and AF.
  • Significant increases in AF concentrations of big ET and ET-1 were observed from mid-trimester to full-term pregnancy (p < 0.01).
  • HPLC analysis revealed ET-1 as a major form in AF, while big ET existed as two components, including endothelin fragment [22-38] (EF).

Conclusions:

  • Three molecular forms of endothelin—ET-1, big ET, and EF—are present in amniotic fluid.
  • Concentrations of ET-1 and big ET are elevated in the AF of women at full-term pregnancy compared to mid-trimester.
