Abstract:
Overexpression of the antiapototic proteins Bcl-2 and Bcl-xL provides a common mechanism through which cancer cells gain a survival advantage and become resistant to conventional chemotherapy. Inhibition of these prosurvival proteins is an attractive strategy for cancer therapy. We recently described the discovery of a selective Bcl-xL antagonist that potentiates the antitumor activity of chemotherapy and radiation. Here we describe the use of structure-guided design to exploit a deep hydrophobic binding pocket on the surface of these proteins to develop the first dual, subnanomolar inhibitors of Bcl-xL and Bcl-2. This study culminated in the identification of 2, which exhibited EC50 values of 8 nM and 30 nM in Bcl-2 and Bcl-xL dependent cells, respectively. Compound 2 demonstrated single agent efficacy against human follicular lymphoma cell lines that overexpress Bcl-2, and efficacy in a murine xenograft model of lymphoma when given both as a single agent and in combination with etoposide.
Insights
Researchers developed novel dual inhibitors targeting Bcl-2 and Bcl-xL proteins, crucial for cancer cell survival. These compounds show promise as single agents and in combination therapy for lymphoma.

- Molecular Biology
- Drug Discovery
Background:
- Overexpression of anti-apoptotic proteins Bcl-2 and Bcl-xL confers cancer cell survival and chemotherapy resistance.
- Inhibiting these prosurvival proteins is a key therapeutic strategy for cancer treatment.
Purpose of the Study:
- To develop novel dual inhibitors of Bcl-2 and Bcl-xL using structure-guided design.
- To identify potent inhibitors that exploit the hydrophobic binding pocket of Bcl-2 and Bcl-xL.
Main Methods:
- Structure-guided drug design targeting the hydrophobic binding pocket of Bcl-2 and Bcl-xL.
- Synthesis and characterization of dual Bcl-2/Bcl-xL inhibitors.
- Evaluation of compound efficacy in Bcl-2 and Bcl-xL dependent cancer cells and in vivo lymphoma models.
Main Results:
- Identification of compound 2, a dual inhibitor with subnanomolar potency (EC50 of 8 nM for Bcl-2 and 30 nM for Bcl-xL).
- Compound 2 demonstrated single-agent efficacy against human follicular lymphoma cell lines overexpressing Bcl-2.
- Compound 2 showed efficacy in a murine xenograft lymphoma model, both as a single agent and in combination with etoposide.
Conclusions:
- Dual inhibition of Bcl-2 and Bcl-xL is achievable with potent small molecules.
- Compound 2 represents a promising therapeutic candidate for Bcl-2-dependent lymphomas.
