Summary

The DC2.4 cell line exhibits characteristics of immature dendritic cells, with low expression of the RelB gene and CD40 molecules, alongside strong phagocytic capacity.

Area of Science:

  • Immunology
  • Cell Biology

Background:

  • Dendritic cells (DCs) are crucial antigen-presenting cells in the immune system.
  • The DC2.4 cell line is a commonly used mouse dendritic cell model.
  • Understanding the phenotype and molecular characteristics of DC2.4 is important for immunological research.

Purpose of the Study:

  • To investigate the relationship between the nuclear factor-kappaB transcription factor RelB gene expression and surface molecules in the DC2.4 cell line.
  • To characterize the phenotype of the DC2.4 cell line.

Main Methods:

  • Cell culture of DC2.4 line in RPMI 1640 medium.
  • Morphological analysis using optical and transmission electron microscopy.
  • Flow cytometry for surface markers (MHC-II, CD86, CD40) and RT-PCR for RelB mRNA expression.

Main Results:

  • DC2.4 cells showed irregular shapes with dendritic processes and cytoplasmic vesicles.
  • Flow cytometry revealed low MHC-II and CD40, but high CD86 expression.
  • Low RelB mRNA expression was detected, similar to immature bone marrow-derived DCs.

Conclusions:

  • DC2.4 cells possess strong phagocytic capacity.
  • Low expression of RelB gene and CD40 suggests an immature dendritic cell phenotype for the DC2.4 line.
  • This characterization aids in the appropriate use of DC2.4 cells in immunological studies.
