Parkinson's disease (PD) involves oxidative stress damaging brain cells. DNA repair enzymes MTH1, OGG1, and MUTYH are upregulated in PD patients' substantia nigra, suggesting their role in mitochondrial DNA repair.

Background:

Parkinson's disease (PD) is a common neurodegenerative disorder.
Oxidative stress and mitochondrial dysfunction contribute to neuronal death in PD's substantia nigra.
Neurons, as post-mitotic cells, possess mechanisms to prevent oxidative DNA damage.

Purpose of the Study:

Investigate the role of DNA repair enzymes in Parkinson's disease.
Determine if specific DNA repair enzymes are upregulated in the substantia nigra of PD patients.
Explore the cooperative function of MTH1, OGG1, and MUTYH in mitochondrial DNA repair within the PD brain.

Main Methods:

Analysis of DNA repairing enzyme expression in human brain tissue.
Quantification of MTH1, OGG1, and MUTYH expression levels.
Comparison of enzyme expression between PD patients and controls.

Main Results:

MTH1, OGG1, and MUTYH enzymes are expressed in the human brain.
All three enzymes (MTH1, OGG1, MUTYH) show increased expression in the substantia nigra of PD patients.
Upregulation suggests a cooperative role in mitochondrial DNA repair.

Conclusions:
