Summary

Quantifying amyloid beta (Abeta) in cerebrospinal fluid (CSF) using direct protein binding arrays (IMAC30) detected elevated Abeta40 levels in Alzheimer's disease (AD). Antibody-based methods did not yield significant differences, suggesting interference.

Background:

  • Amyloid beta (Abeta) is a key component of senile plaques in Alzheimer's disease (AD).
  • Soluble Abeta oligomers are increasingly implicated in AD pathogenesis.

Purpose of the Study:

  • To compare three methods for quantifying Abeta40 in CSF.
  • To evaluate the impact of different quantification strategies on AD biomarker detection.

Main Methods:

  • Compared ELISA, antibody-based SELDI-TOF, and direct binding SELDI-TOF (IMAC30) for Abeta40 quantification.
  • Analyzed cerebrospinal fluid (CSF) samples.

Main Results:

  • Only SELDI-TOF with IMAC30 arrays showed significantly elevated CSF Abeta40 in AD patients (127% of controls).
  • Antibody-based methods did not detect significant differences in Abeta40 levels between AD patients and controls.

Conclusions:

  • CSF Abeta40 measurement is method-dependent.
  • Antibody-based detection may be affected by Abeta oligomerization or binding to carrier proteins, potentially masking epitopes.
