Summary

Matrix metalloproteinase inhibitors show promise for treating diseases beyond arthritis and cancer. Shorter, targeted treatment durations may reduce dose-limiting side effects, making MMP inhibition a viable therapeutic strategy.

Role of Matrix Metalloproteases in Degradation of ECM

Matrix metalloproteases (MMPs) are enzymes involved in the hydrolysis of proteins and glycoproteins of the extracellular matrix. MMPs are essential for the migration and proliferation of cells through the dense matrix network, throughout embryonic development, and throughout morphogenesis. The first MMP activity discovered was a collagenase in a tadpole's tail undergoing metamorphosis. The active collagen deposition and modifications lead to the morphogenesis of tadpoles into the adult body.

Area of Science:

  • Biochemistry
  • Pharmacology
  • Enzymology

Background:

  • Matrix metalloproteinases (MMPs) are a family of enzymes targeted by pharmaceuticals for over two decades.
  • Despite extensive drug development, no MMP inhibitors have succeeded in clinical trials due to dose-limiting side effects.
  • MMP activity is implicated in the pathophysiology of various diseases, presenting potential therapeutic opportunities.

Purpose of the Study:

  • To explore alternative clinical settings where MMP inhibition is a potentially valid therapeutic strategy.
  • To review evidence from animal models supporting MMP inhibition.
  • To identify scenarios where reduced drug exposure may mitigate side effects.

Main Methods:

  • Review of existing literature on MMPs in disease pathophysiology.

Bacterial Expression and Purification of Human Matrix Metalloproteinase-3 using Affinity Chromatography

Published on: March 30, 2022

  • Analysis of data from animal models investigating MMP inhibition.
  • Evaluation of potential therapeutic strategies focusing on reduced treatment duration and systemic exposure.
  • Main Results:

    • Strong evidence suggests MMP involvement in diseases beyond arthritis and cancer.
    • Animal models demonstrate the efficacy of MMP inhibition in specific disease contexts.
    • Targeted, short-term, or localized MMP inhibition may circumvent historical side-effect issues.

    Conclusions:

    • MMP inhibition remains a promising therapeutic avenue, particularly in diseases with strong MMP involvement.
    • Alternative administration strategies, such as reduced duration or systemic exposure, can overcome previous clinical trial limitations.
    • Further investigation into specific disease settings and optimized MMP inhibitor therapy is warranted.
