Southern Blot

Agarose gel electrophoresis is very useful in separating DNA fragments by size. Running a DNA ladder containing fragments of the known length alongside the sample helps determine the approximate length of the sample DNA fragments. However, additional steps are needed to verify the sequence identity of the sample DNA fragments.
Denatured DNA fragments must be transferred onto a carrier membrane from the gel to make it accessible to a probe - a small ssDNA fragment complementary to the target DNA...

Modern Molecular Taxonomy

Advancements in molecular biology have revolutionized the identification and characterization of bacteria, with multiple methods leveraging DNA sequencing for enhanced precision. As sequencing technologies improve and costs decline, these approaches are increasingly used in clinical, environmental, and evolutionary studies.Multilocus Sequence Typing (MLST) examines several housekeeping genes, essential chromosomal genes encoding cellular functions, to distinguish strains. Labeling DNA Probes03:31

Labeling DNA Probes

DNA probes are fragments of DNA labeled with a reporter tag to enable their detection or purification. The resulting labeled DNA probes can then hybridize to target nucleic acid sequences through complementary base-pairing, and may be used to recover or identify these regions.
Radioisotopes, fluorophores, or small molecule binding partners like biotin or digoxigenin, are the most widely used reporter tags for labeling DNA probes. These labels can be attached to the probe DNA molecule via...

Summary

A new microarray assay accurately identifies single nucleotide polymorphisms (SNPs) in olive varieties. This method, using ligation detection reaction-universal array (LDR-UA), effectively distinguishes 49 olive types for olive oil production.

Area of Science:

  • Agricultural Science
  • Genetics
  • Molecular Biology

Background:

  • Assessing the origin and authenticity of food products is crucial.
  • Molecular methods offer precise tools for verifying raw materials and processed goods.

Purpose of the Study:

  • To apply a microarray-based assay for identifying single nucleotide polymorphisms (SNPs) in olive cultivars.
  • To validate the accuracy of the assay against direct sequencing.
  • To determine the efficacy of a panel of SNPs in discriminating olive varieties.

Main Methods:

  • Utilized a ligation detection reaction-universal array (LDR-UA) assay.
  • Identified 17 single nucleotide polymorphisms (SNPs) in olive genomic sequences.
  • Amplified olive DNA from fresh leaves using PCR.
  • Detected alleles via fluorescent hybridization on a universal array.

Main Results:

  • The LDR-UA assay demonstrated full agreement with direct sequencing for genotype calls.
  • A panel of 17 SNPs was sufficient to differentiate 49 distinct olive varieties.
  • The method is effective for authenticating olive cultivars used in olive oil production.

Conclusions:

  • The LDR-UA assay is a reliable and accurate method for SNP identification in olive cultivars.
  • This molecular tool aids in the discrimination of olive varieties, particularly those significant for olive oil production in the Mediterranean region.
  • The study provides a valuable method for ensuring the authenticity and traceability of olive products.
