  • Determination of absolute configurations via optical rotation and Circular Dichroism (CD) spectroscopy.
  • In vitro assays for cytotoxicity, MDR reversal (calcein accumulation), and anti-inflammatory activity (ICAM-1 induction).

    • Six new lignans (compounds 1-6) and 14 known compounds were identified.
    • Compound 3 displayed significant cancer cell growth inhibition against VA-13 and HepG2 cell lines (IC50 values 5.3 and 13.2 μg/mL, respectively).
    • Compound 3 showed superior MDR reversal activity in 2780AD cells compared to verapamil.
    • Compound 6 exhibited anti-inflammatory activity by inhibiting IL-1α and TNF-α-induced ICAM-1 expression.

    Conclusions:

    • Peperomia duclouxii is a rich source of structurally diverse lignans.
    • The novel lignans possess promising cytotoxic and MDR reversal properties.
    • Compound 6 demonstrates potential as an anti-inflammatory agent.
