Summary
Genetic variations in the interleukin-12B (IL-12B) gene are linked to psoriasis vulgaris (PV) susceptibility in Taiwanese Chinese. However, HLA-Cw*0602 showed a stronger association, particularly with early-onset PV.
Area of Science:
- Genetics
- Immunology
- Dermatology
Background:
- Cytokine gene polymorphisms are implicated in psoriasis susceptibility.
- Understanding genetic factors is crucial for elucidating psoriasis pathogenesis.
Purpose of the Study:
- To investigate the association between cytokine gene polymorphisms and psoriasis development in Taiwanese Chinese patients.
- To assess the role of specific genetic markers in psoriasis vulgaris (PV) and psoriatic arthritis (PsA).
Main Methods:
- Analyzed DNA from 170 PV patients, 102 PsA patients, and 210 controls using direct sequencing and microsatellite genotyping.
- Examined 28 polymorphisms in 11 cytokine genes (e.g., IL-1, IL-4, IL-10, TNF-alpha).
- Determined genotypes for HLA-Cw*0602, killer cell immunoglobulin-like receptor (KIR) genes, and major histocompatibility complex class I chain-related gene A (MICA) in PsA patients.
Main Results:
- The +4496G allele of the IL-12B gene was more frequent in PV patients (59.4%) compared to controls (49.3%).
- HLA-Cw*0602 allele frequency was significantly higher in early-onset PV patients (25.3%) versus controls (4.8%).
- No significant associations were found for other analyzed cytokine genes, KIR genes, or MICA with PV or PsA, nor with disease onset, gender, or arthritis characteristics.
Conclusions:
- IL-12B gene polymorphism is a risk factor for PV in the Chinese population in Taiwan, though less impactful than HLA-Cw*0602.
- HLA-Cw*0602 is a significant risk factor for early-onset PV.
- Specific polymorphisms in IL-12B and HLA-Cw*0602 are associated with psoriasis susceptibility, while other tested cytokine gene polymorphisms, KIR, and MICA are not strongly linked to PV or PsA in this cohort.

Single Nucleotide Polymorphisms-SNPs
A single nucleotide polymorphism or SNP is a single nucleotide variation at a specific genomic position in a large population. It is the most prevalent type of sequence variation found in the human genome. Point mutations that occur in more than 1% of the population qualify as SNPs. These are present once every 1000 nucleotides on an average in the human genome. Replacement of a purine with another purine (A/G) or a pyrimidine with another pyrimidine (C/T) is known as a transition. In contrast,...
Pharmacogenetic Phenotypes: Alterations in Pharmacokinetics, Drug Targets and Biologic Milieu
Genetic variations significantly influence drug response through pharmacokinetics, receptor interactions, and biologic milieu modifications. Pharmacokinetic alterations impact drug metabolism and clearance, affecting efficacy and toxicity. Variants in drug-metabolizing enzymes, such as CYP2C9 and CYP2C19, alter drug activation and elimination. For example, CYP2C9 loss-of-function variants require lower warfarin doses to prevent excessive bleeding, while CYP2C19 variants reduce clopidogrel...
Pharmacogenetics of Drug Targets: β₂-Adrenergic Receptors, Apo E, Thymidylate Synthase
Genetic polymorphisms in drug targets have emerged as critical determinants of interindividual variability in drug response and toxicity. Pharmacogenomic investigations increasingly focus on identifying these variations to personalize and optimize therapeutic interventions. A drug target may be a receptor, enzyme, or signaling protein involved in pharmacologic responses or disease-related pathways. While early pharmacogenetic studies focused primarily on drug metabolism, current research...
Pharmacogenetics of Phase I Enzymes: Cytochrome P450 Isozymes
Cytochrome P450 (CYP450) enzymes are a superfamily of heme-containing monooxygenases that play a pivotal role in Phase I drug metabolism by catalyzing oxidation and reduction reactions. These enzymes transform lipophilic xenobiotics into more hydrophilic metabolites, facilitating subsequent Phase II conjugation and eventual excretion. The CYP450 family is classified into families (e.g., CYP1–CYP3) and subfamilies (e.g., CYP2A, CYP2C), based on amino acid sequence homology.CYP450 isoenzymes,...
Comparing Copy Number Variations and SNPs
Sequencing of the human genome has opened up several best-kept secrets of the genome. Scientists have identified thousands of genome variations that exist within a population. These variations can be a single nucleotide or a larger chromosomal variation.
Copy number variations or CNVs are the structural variations that cover more than 1kb of DNA sequence. The single nucleotide polymorphism (SNP), on the other hand, is a single nucleotide change or a point mutation that is found in more than 1%...

The JAK-STAT Signaling Pathway
Several cytokine receptors have tightly bound Janus kinase or JAK proteins attached at their cytosolic tail. Small signaling molecules such as cytokines, growth hormones, or prolactins bind to the cytokine receptors and initiate their dimerization. The dimerization brings the cytosolic JAKs together that trans-phosphorylate and activates each other. The activated JAKs now phosphorylate cytosolic tails of the cytokine receptors, which serve as binding sites for adaptor proteins such as SH2...
