Transgenic Plants02:50

Transgenic Plants

Recombinant DNA technology called transgenesis is often used to add a foreign gene or remove a detrimental gene from an organism. Such genetically modified organisms are called transgenic organisms.
The first-ever transgenic plant was a tobacco plant developed in 1983 that showed resistance against the tobacco mosaic virus. Since then, many transgenic plants have been developed and commercialized for improving the agricultural, ornamental, and horticultural value of a crop plant. Transgenic...

Area of Science:

  • Plant Biotechnology
  • Genetics and Molecular Biology
  • Agricultural Science

Background:

  • Agrobacterium-mediated transformation is crucial for rice functional genomics.
  • Current methods are limited to japonica rice varieties with high regeneration potential.
  • Elite japonica and most indica rice varieties exhibit low transformation efficiencies due to regeneration difficulties.

Purpose of the Study:

  • To develop an improved Agrobacterium-mediated rice transformation protocol.
  • To enhance transformation efficiency across diverse rice genotypes, including recalcitrant varieties.
  • To facilitate high-throughput functional analysis in a broader range of rice.

Main Methods:

  • Utilized an optimized Agrobacterium-mediated transformation approach.
  • Applied the method to both easily cultured and elite japonica rice varieties.
  • Focused on improving regeneration and transformation efficiency.

Main Results:

  • Achieved successful Agrobacterium-mediated transformation in elite japonica rice.
  • Obtained transgenic rice plants within a 2-3 month timeframe.
  • Reported transformation frequencies ranging from 30-50% for both easily cultured and recalcitrant varieties.

Conclusions:

  • The developed method broadens the applicability of Agrobacterium-mediated rice transformation.
  • This protocol overcomes regeneration limitations in elite japonica rice.
  • Enables efficient genetic modification for functional studies in diverse rice germplasms.
