Background:

  • Tuberculosis (TB) is a major infectious disease in India, with diagnosis challenged by slow Mycobacterium tuberculosis growth.
  • Conventional TB diagnostic tools require significant time due to the slow growth rate of Mycobacterium tuberculosis.

Purpose of the Study:

  • To compare the diagnostic efficacy of the novel Fast Plaque TB assay against conventional methods for tuberculosis diagnosis.
  • To evaluate the sensitivity and specificity of the phage assay for detecting Mycobacterium tuberculosis in clinical samples.

Main Methods:

  • Two hundred and twelve clinically suspected TB cases were enrolled.
  • Three consecutive sputum samples were collected for Ziehl-Neelsen (ZN) staining, culture on LJ media, and phage assay.
  • Phage assay was performed to detect Mycobacterium tuberculosis.

Main Results:

  • The phage assay showed a sensitivity of 94.34% and specificity of 93.88% compared to AFB smear.
  • Compared to LJ media culture, the phage assay demonstrated 92.86% sensitivity and 97.83% specificity.
  • All 8 Non-Tubercular Mycobacteria tested were negative by the Fast Plaque Assay.

Conclusions:

  • The Fast Plaque TB assay is a rapid, reliable, and cost-effective method for diagnosing pulmonary tuberculosis.
  • The phage assay provides a significant improvement over conventional diagnostic methods for TB.
