Purpose of the Study:

  • To investigate and compare the ultrastructure of spermatozoa across selected chironomid, Culicoides, and Odagmia pontina species.
  • To identify conserved and divergent features in sperm morphology within the Chironomoidea superfamily.

Main Methods:

  • Transmission electron microscopy (TEM) was employed to examine the fine structure of spermatozoa.
  • Comparative analysis of sperm components, including acrosome, axoneme, and centriole adjunct, was performed.

Main Results:

  • A synapomorphic feature of nine kidney-shaped structures forming the centriole adjunct was identified in chironomid species.
  • All Chironomoidea members exhibit a mono-layered acrosome and a flagellar axoneme with accessory tubules containing 15 protofilaments.
  • A 9+9+2 axoneme pattern was common, with a rare 9+9+0 variation observed in an unidentified chironomid species lacking central complex and spokes.
  • Sperm motility was preserved across all examined species, and spermatozoa showed signs of completing maturation within the deferent ducts.

Conclusions:

  • The centriole adjunct structure is a significant synapomorphic feature for chironomid species.
  • Despite axonemal variations, sperm motility is a conserved trait in these insect groups.
  • Spermatozoa maturation is a dynamic process occurring during transit through the deferent ducts.
