This study investigated the chemical stability of lipids in Solid Lipid Nanoparticles (SLN). Triglyceride-based SLN formulations exhibited excellent stability, with Dynasan 118 showing the best performance over two years.
Area of Science:
- Pharmaceutical Technology
- Materials Science
- Physical Chemistry
Background:
- Lipids are crucial excipients in Solid Lipid Nanoparticles (SLN) formulation.
- Understanding lipid chemical stability within SLN is vital for drug delivery applications.
- Excipient stability directly impacts the shelf-life and efficacy of nanoparticle-based therapeutics.
Purpose of the Study:
- To evaluate the chemical stability of various lipids used as excipients in SLN.
- To determine the influence of lipid type and surfactant composition on SLN stability.
- To assess the impact of the SLN production process on lipid excipient integrity.
Main Methods:
- Formulation of SLN using different lipids and surfactant systems via hot homogenization.
- Characterization of SLN using photon correlation spectroscopy, laser diffractometry, zeta potential, and differential scanning calorimetry.
- Chemical stability assessment of lipid excipients using gas chromatography (GC) after lipid extraction.
Main Results:
- The SLN production process did not compromise the chemical stability of the lipid matrix.
- SLN formulations based on triglycerides demonstrated minimal lipid decomposition at 25°C.
- Dynasan 118 exhibited superior chemical stability, with less than 4% degradation over two years.
Conclusions:
- Triglyceride-based lipids are highly stable excipients for SLN production.
- Dynasan 118 is a promising lipid excipient for long-term stable SLN formulations.
- SLN manufacturing via hot homogenization preserves the chemical integrity of lipid excipients.
