Curcumin analogues, oxazoles and pyrazoles, were synthesized to improve gamma-secretase inhibition by reducing metal chelation. These novel compounds effectively inhibit gamma-secretase, showing potent activity in the low micromolar range.
Area of Science:
- Medicinal Chemistry
- Organic Synthesis
- Neuroscience
Background:
- Curcumin exhibits metal chelation properties, which can interfere with its biological activity.
- Gamma-secretase is a key enzyme implicated in Alzheimer's disease pathogenesis.
- Modifying curcumin's structure is a strategy to enhance its therapeutic potential.
Purpose of the Study:
- To synthesize curcumin-derived oxazoles and pyrazoles.
- To minimize the metal chelation properties of curcumin.
- To develop potent gamma-secretase inhibitors.
Main Methods:
- Synthesis of curcumin analogue oxazoles and pyrazoles.
- Evaluation of metal chelation properties.
- Assay of gamma-secretase inhibition activity.
Main Results:
- Curcumin-derived oxazoles and pyrazoles were successfully synthesized.
- The synthesized compounds exhibited reduced metal chelation compared to curcumin.
- Compounds 4a-i demonstrated potent gamma-secretase inhibition in the low micromolar range.
Conclusions:
- Replacement of the 1,3-dicarbonyl moiety with isosteric heterocycles yields potent gamma-secretase inhibitors.
- These novel curcumin analogues offer a promising therapeutic strategy for conditions involving gamma-secretase.
- Reduced rotational freedom and absence of stereoisomers contribute to enhanced inhibitory activity.

Allosteric Proteins-ATCase
Aspartate transcarbamoylase (ATCase) is a cytosolic enzyme that catalyzes the condensation of L-aspartate and carbamoyl phosphate to N-carbamoyl-L-aspartate. This reaction is the first step in pyrimidine biosynthesis. UTP and CTP, the end products of the pyrimidine synthesis pathway,...
