Background:

  • The surface-connected open canalicular system (OCS) in human platelets is crucial for internal transport and secretion.

Purpose of the Study:

  • To investigate whether the OCS of human platelets can simultaneously transport substances into the cell and discharge alpha granule contents.
  • To determine if the OCS serves as a common pathway for both platelet functions.

Main Methods:

  • Human platelets were stimulated with thrombin in the presence of fibrinogen-coupled gold nanoparticles (Fgn/Au).
  • Immunogold labeling with anti-fibrinogen antibodies was used to track endogenous and exogenous fibrinogen.
  • Electron microscopy and immunocytochemistry were employed to visualize particle localization within platelet structures.

Main Results:

  • Thrombin stimulation led to Fgn/Au particle uptake into the OCS channels.
  • Endogenous fibrinogen was observed in alpha granules and subsequently in the OCS.
