Interleukin-21 (IL-21) differentially affects human natural killer (NK) cell subpopulations. IL-21 promotes CD56(bright) NK cell proliferation and enhances CD56(dim) NK cell cytotoxicity, with additive effects when combined with IL-2.

Background:

  • Interleukin-21 (IL-21) is a cytokine impacting various immune cells.
  • Human natural killer (NK) cells exist as distinct subpopulations: CD56(dim) and CD56(bright).

Purpose of the Study:

  • To investigate the differential effects of IL-21 on human CD56(dim) and CD56(bright) NK cell subpopulations.
  • To compare IL-21's functional impact with IL-2 and their combined effects on NK cells.

Main Methods:

  • Functional assays on isolated CD56(dim) and CD56(bright) NK cells stimulated with IL-21 and/or IL-2.
  • Analysis of cell proliferation, cytotoxicity, cytokine production (IFN-γ, TNF-α), and STAT protein phosphorylation (STAT1, STAT3, STAT5).

Main Results:

  • IL-21 significantly induced proliferation in CD56(bright) NK cells.
  • IL-21 preferentially augmented cytotoxicity in CD56(dim) NK cells.
  • IL-21 induced STAT1 and STAT3 phosphorylation in both subsets, while IL-2 primarily phosphorylated STAT5 in CD56(bright) cells. Combined IL-2 and IL-21 showed additive effects on STAT phosphorylation.

Conclusions:

  • IL-21 exerts distinct functional effects on human NK cell subpopulations.
  • Differential STAT signaling pathways are activated by IL-2 and IL-21 in NK cells.
  • IL-2 and IL-21 can act additively, influencing NK cell functions and signaling.
