Enhanced lymphatic drainage, measured by lymph node capacity (LNcap), shows a protective effect against inflammatory arthritis progression. Lower LNcap in TNF-Tg mice correlated with increased arthritis severity, indicating its prognostic value.

Area of Science:

  • Immunology
  • Medical Imaging
  • Rheumatology

Background:

  • Inflammatory arthritis leads to erosion and tissue necrosis.
  • Prognostic factors for arthritis initiation and severity are largely unknown.
  • Contrast-enhanced (CE)-magnetic resonance imaging (MRI) is a novel tool for studying arthritis pathogenesis.

Purpose of the Study:

  • To develop CE-MRI for mouse knee to identify prognostic factors in inflammatory arthritis.
  • To assess synovitis and draining lymph node (LN) function in TNF-Tg mice.
  • To investigate the relationship between LN function and arthritis severity.

Main Methods:

  • Developed dynamic CE-MRI for mouse knee imaging.
  • Quantified lymph node (LN) volume and enhancement for LN capacity (LNcap) measurement.

  • Correlated LNcap with synovial volume and arthritis progression in TNF-Tg and wild-type (WT) mice.
  • Main Results:

    • TNF-Tg mice exhibited significantly larger LNs with greater enhancement compared to WT mice.
    • LNcap was 15-fold higher in TNF-Tg mice, decreasing significantly with anti-TNF therapy.
    • A negative correlation between LNcap and synovial volume indicated that lower LNcap accelerates arthritis progression.

    Conclusions:

    • Enhanced lymphatic drainage, reflected by increased LNcap, plays a protective role in inflammatory arthritis.
    • LNcap is a potential prognostic biomarker for predicting arthritis severity.
    • CE-MRI provides valuable insights into the pathogenesis and potential therapeutic targets of inflammatory arthritis.
