This study characterizes a new copper(II) complex of the antihypertensive drug Losartan. The Losartan copper(II) complex exhibits antioxidant properties and differential toxicity to tumoral cells compared to Losartan alone.
Area of Science:
- Pharmaceutical Chemistry
- Coordination Chemistry
- Biomedical Science
Background:
- Losartan is a widely used antihypertensive drug.
- Copper complexes can exhibit unique biological activities.
- Understanding Losartan's coordination chemistry is important for novel drug development.
Purpose of the Study:
- To synthesize and characterize a novel copper(II) complex with Losartan.
- To investigate the antioxidant and cytotoxic properties of the Losartan-copper(II) complex.
- To evaluate the complex's effects on osteoblast-like cells.
Main Methods:
- Vibrational spectroscopy (FTIR and Raman) for structural analysis.
- Electron Paramagnetic Resonance (EPR) spectroscopy for impurity detection.
- In vitro cell culture assays to assess proliferation, morphology, and toxicity.
Main Results:
- The copper(II) complex was synthesized and characterized, revealing a dimeric structure with a tetragonal distorted metal center and a monomeric impurity.
- The Losartan ligand coordinated to copper(II) via the tetrazolate nitrogen atoms.
- The complex demonstrated superoxide dismutase mimetic activity and selective toxicity towards tumoral cells.
Conclusions:
- The novel Losartan-copper(II) complex possesses distinct structural and biological properties.
- The complex shows potential as an antioxidant agent with enhanced cytotoxic effects on tumoral cells.
- Further research into Losartan-metal complexes may yield new therapeutic agents.
