The factor for inversion stimulation (Fis) protein inhibits IS2 transposition in Escherichia coli by binding to key DNA sites. Fis reduces IS2 gene transcription and blocks transposase binding, significantly lowering transposition frequency.
Area of Science:
- Molecular Biology
- Microbiology
- Genetics
Background:
- IS2 is a mobile genetic element in Escherichia coli.
- The factor for inversion stimulation (Fis) protein is known to regulate various DNA processes.
Purpose of the Study:
- To investigate the role of Fis protein in regulating IS2 transposition.
- To elucidate the mechanisms by which Fis affects IS2 transposition.
Main Methods:
- Transposition assays in wild-type and fis-mutant Escherichia coli.
- Gel retardation and DNase I footprinting to analyze Fis DNA binding.
- Luciferase reporter assays to measure IS2 promoter activity.
Main Results:
- IS2 transposition frequency was 64-fold lower in wild-type E. coli compared to fis-mutants.
- Fis protein binds to specific sites within the IS2 major promoter and at the right terminal repeat.
- Fis binding reduced transcription from the IS2 major promoter by approximately sixfold.
- Fis binds to the same region as the IS2 transposase at the right terminal repeat.
Conclusions:
- Fis protein inhibits IS2 transposition through a dual mechanism.
- Fis represses IS2 gene transcription by binding to the promoter region.
- Fis blocks IS2 transposition by interfering with IS2 transposase binding to the terminal repeat.
