Summary

The DSD-1 epitope and phosphacan core protein show distinct expression patterns during mouse visual system development. Phosphacan is expressed by neurons, suggesting a role in retinal development and neuronal differentiation.

Area of Science:

  • Neuroscience
  • Developmental Biology
  • Cell Biology

Background:

  • Receptor-protein-tyrosine-phosphatase-beta/zeta (RPTPbeta/zeta) has splice variants, including DSD-1-PG/phosphacan.
  • RPTPbeta/zeta exists in long and short isoforms, expressed by glial cells.
  • Secreted phosphacan isoforms bind extracellular matrix and influence neuronal differentiation.

Purpose of the Study:

  • To investigate the expression patterns of the DSD-1 epitope and phosphacan isoforms in the developing mouse visual system.
  • To determine the cellular localization and developmental timing of these molecules in the retina.
  • To explore the role of phosphacan/RPTPbeta isoforms in retinal development.

Main Methods:

  • Immunohistochemistry to detect DSD-1 epitope and phosphacan core protein.

Temporal Ordering of Dynamic Expression Data from Detailed Spatial Expression Maps

Published on: February 9, 2017

  • Reverse-transcriptase polymerase chain reaction (RT-PCR) to analyze gene expression levels.
  • In situ hybridization to determine cellular expression patterns.
  • Main Results:

    • DSD-1 epitope appears at E13, peaks at P6, and is downregulated by adolescence, localized to nerve fiber and inner plexiform layers.
    • Phosphacan core protein is detectable from E12, peaks at P14, and persists into adulthood, with broader retinal layer distribution.
    • RT-PCR and in situ hybridization suggest shifts in phosphacan/RPTPbeta isoform expression during development, with retinal neurons expressing at least one isoform.

    Conclusions:

    • DSD-1 epitope and phosphacan core protein exhibit differential temporal and spatial expression during mouse retinal development.
    • Phosphacan/RPTPbeta isoforms show developmental stage-specific expression patterns.
    • Evidence suggests that phosphacan/RPTPbeta isoforms are expressed by retinal neurons, potentially mediating neuronal differentiation and visual system development.
