Summary

Flomoxef (FMOX), an oxacephem antibiotic, shows promising pharmacokinetic and clinical effects in neonates. This study found FMOX to be safe and effective, even against resistant bacteria.

Background:

  • Parenteral oxacephem antibiotics are crucial in treating neonatal infections.
  • Flomoxef (FMOX) is a newer antibiotic in this class requiring pharmacokinetic and clinical evaluation in neonates.

Observation:

  • Pharmacokinetic studies measured serum concentrations and urinary recovery rates of FMOX at different doses (20 mg/kg and 40 mg/kg).
  • Clinical effects were observed in four neonates with various infections, including sepsis and methicillin-resistant Staphylococcus aureus (MRSA).

Findings:

  • Mean peak serum concentrations of FMOX were 33.3 µg/ml (20 mg/kg) and 68.9 µg/ml (40 mg/kg).
  • Mean 6-hour urinary recovery rates were 35.2% (20 mg/kg) and 48.3% (40 mg/kg).
  • Excellent clinical outcomes were achieved in 3 out of 4 neonates, including an MRSA infection, with doses of 20-30 mg/kg.
